A Clinical Study from Jazz Pharmaceuticals for Adults Who Have Been Diagnosed with HER2-Positive Advanced Gastroesophageal Adenocarcinoma (GEA)

This study is evaluating the safety and efficacy of two investigational medicines called zanidatamab and tislelizumab for the treatment of HER2-positive advanced or metastatic (which means the cancer has spread to other parts of the body) gastroesophageal adenocarcinoma (GEA), including stomach cancer, gastroesophageal junction cancer, and esophagus cancer. The potential clinical effectiveness of zanidatamab is still being tested and it is not currently approved for treatment of any disease state in any market. The purpose of this page is to inform viewers of the current clinical trial.

Trial Purpose

The purpose of the HERIZON GEA-301 study is to find out if zanidatamab combined with chemotherapy, with or without tislelizumab, works better than a current standard therapy: trastuzumab combined with chemotherapy.

What is gastroesophageal adenocarcinoma (GEA)?

Gastroesophageal adenocarcinoma (GEA) is a type of cancer that develops in the cells of the mucous glands in the lining of the esophagus or stomach. Adenocarcinoma is one of the two most common types of esophageal cancer (esophageal cancer refers to malignancies that develop in the esophagus, the muscular tube that connects the throat to the stomach) and can also occur in the junction where the esophagus meets the stomach, known as the gastroesophageal junction, as well as in the stomach.

GEA typically develops over time, often starting as precancerous changes in the cells of the esophagus or stomach. Risk factors for GEA include gastroesophageal reflux disease (GERD), Barrett’s esophagus (a condition in which the cells lining the lower esophagus change in response to repeated exposure to stomach acid), H. pylori infection, obesity, smoking, and a diet high in processed meats or low in fruits and vegetables.

Symptoms of GEA may include difficulty swallowing, chest pain, unintended weight loss, indigestion or heartburn, vomiting, and fatigue.

Treatment options for GEA depend on the stage of the cancer and may include surgery, chemotherapy, radiation therapy, molecularly targeted therapy, and immunotherapy. Early detection and treatment offer the best chance of a favorable outcome.

What are the drugs being investigated?

This open-label, randomized Phase 3 clinical trial will study two investigational therapies (zanidatamab and tislelizumab) for HER2-positive GEA.

Zanidatamab is a drug that is being explored as an investigational therapy. Zanidatamab is designed to target specific forms of cancer that express a protein called HER2. A doctor can test if a patient’s cancer expresses HER2. Zanidatamab is believed to work by blocking the HER2 signal on cancer cells while removing the HER2 protein from the cell surface. HER2 signaling tells cancer cells to survive, grow, and multiply; blocking the growth signal and removing HER2 protein from the cell surface may treat tumors. In addition, zanidatamab appears to activate the immune system to attack tumor cells, which may also help to treat a patient’s HER2-expressing cancer.

Tislelizumab (Tevimbra®) is a drug that is approved for the treatment of some cancers and is also being explored in combination with other treatments as an investigational therapy for the treatment of HER2-positive GEA and other cancers. Tislelizumab belongs to a class of medicines known as checkpoint inhibitors and works by blocking the PD-1 pathway to help prevent cancer cells from hiding from the immune system, helping the body’s immune system fight cancer.

Trastuzumab (Herceptin®) is a drug that, when combined with chemotherapy, has been approved in many countries to treat HER2-positive gastric and gastroesophageal cancers. Although not specifically approved for HER2-positive esophageal adenocarcinomas, it is included in global treatment guidelines for this disease.

This study will evaluate and compare the efficacy and safety of zanidatamab plus chemotherapy with or without tislelizumab to the standard of care (Herceptin plus chemotherapy) as first-line treatment for patients with advanced/metastatic HER2-positive GEA.

Other Information

The trial is designed to have approximately 900 patients with HER2-positive GEA cancer participate. Recruitment is ongoing at around 300 study sites in the following countries: Argentina, Australia, Belgium, Brazil, Canada, Chile, China, Estonia, France, Georgia, Germany, Greece, Guatemala, India, Ireland, Italy, Japan, Kenya, Malaysia, Mexico, Netherlands, Poland, Portugal, Republic of Korea, Romania, Serbia, Singapore, South Africa, Spain, Taiwan, Thailand, Turkey, Uganda, and United Kingdom.

Participants in the study will receive the study medicines as well as all study-related visits, tests, and assessments, at no cost. Caregivers of participants may also be reimbursed for some study-related expenses, such as costs associated with travel and hotels.
